Tyrosine kinase activity in the epidermal growth factor receptor.
I have used the baculovirus expression system to over-express the complete
intracellular tyrosine kinase domain and a carboxy terminal truncation of the
EGF receptor as a soluble protein with the ultimate goal of producing
sufficient quantity of protein for crystallographic analysis. The recombinant
proteins have been purified to homogeneity by immunoaffinity chromatography
and enzymatically characterized in vitro. The activity of the tyrosine kinase
domain has been compared to the EGF-activated EGF receptor, and the effect of
various activating molecules has been studied.
